Schizophrenia - associated Chromosome I I q 2 I Translocation : Identification of Flanking Markers and Development of Chromosome I I q Fragment Hybrids as Cloning and Mapping Resources

چکیده
Genetic linkage, molecular analysis, and in situ hybridization have identified TYR and D11S388 as markers flanking the chromosome 11 breakpoint in a large pedigree where a balanced translocation, t(1;11)(q43;q21), segregates with schizophrenia and related affective disorders. Somatic cell hybrids, separating the two translocation chromosomes from each other and from the normal homologues, have been produced with the aid of immunomagnetic sorting for chromosome 1and chromosome 11-encoded cell-surface antigens. The genes for two of these antigens map on either side of the llq breakpoint. Immunomagnetic bead sorting was also used to isolate two stable X-irradiation hybrids for each cell-surface antigen. Each hybrid carries only chromosome 11 fragments. Translocation and X-irradiation hybrids were analyzed, mainly by PCR, for the presence of 19 chromosome 11 and 4 chromosome 1 markers. Ten newly designed primers are reported. The X-irradiation hybrids were also studied cytogenetically, for human DNA content, by in situ Cotd DNA hybridization and by painting the Alu-PCR products from these four lines back onto normal human metaphases. The generation of the translocation hybrids and of the chromosome 1lq fragment hybrids is a necessary preliminary to determining whether a schizophrenia-predisposition gene SCZD2 is encoded at this site.

I-34: NRY Haplotype Analysis: towards A Better Understanding of The Genetic Basis of Spermatogenic Failure
It has been established that the Y chromosome carries genes required for spermatogenesis and male fertility. For many decades worldwide screening for gene identification has been conducted in research laboratories. However, it has been a difficult process in identifying such genes (i.e. causative mutations) which could explain the phenotypic variation and could be potentially used as markers fo...
متن کاملI-38: Chromosome Instability in The Cleavage Stage Embryo
Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...
متن کاملI-3: Human Y Chromosome Proteome Project 2012 Update
The Human Genome Project has generated a blueprint for the approximately 20,300 gene-encoded proteins potentially active in any of 230 cell types that make up the human body (human proteome). However, based on the UniProtKB/Swiss-Prot database content, about 6000 of at the protein level; for many others, there is very little information related to protein function, abundance, subcellular locali...
متن کاملLeft I-quotients of band of right cancellative monoids
Let $Q$ be an inverse semigroup. A subsemigroup $S$ of $Q$ is a left I-order in $Q$ and $Q$ is a semigroup of left I-quotients of $S$ if every element $qin Q$ can be written as $q=a^{-1}b$ for some $a,bin S$. If we insist on $a$ and $b$ being $er$-related in $Q$, then we say that $S$ is straight in $Q$. We characterize semigroups which are left I-quotients of left regular bands of right cancell...
متن کاملI-49: Human Y Chromosome ProteomeProject
The success of the Human Genome Project (HGP) has provided a blueprint for the approximately 20,000 gene-encoded proteins potentially active in all of the hundreds of cell types that make up the human body. Yet we still have limited knowledge about a majority of the gene-encoded proteins which are the “building blocks of life” and “cellular machinery”. It is estimated that for nearly half of th...
